Applications are invited for the post of Junior Research Fellow (JRF) in a SERB-funded research project entitled "A Divergent Strategy for the Asymmetric Total Synthesis of Biologically Active Tetrahydroquinoline and Indole Terpenoids Aspernomine and Tubingensin A" under the supervision of Dr. Santanu Ghosh, Department of Chemistry, School of Natural Sciences, Shiv Nadar Institution of Eminence, Delhi NCR.

Project Synopsis
The rich structural diversity of the tetrahydroquinoloine and indole-based diterpene alkaloids are valuable sources of traditional drugs, pharmaceuticals, and ideas for developing new synthetic strategies and methodologies in chemical synthesis. A study of fungal sclerotia of the genus Aspergillus led to the isolation of several biologically active secondary metabolites, including the complex indole- and tetrahydroquinoline-based diterpenoids. Such as anominine, aflavinine, 17-hydroxyeujindole, tubingensin A, and aspernomine have demonstrated various biological activities such as anti-insecticidal activity, significant cytotoxicity against three human solid tumor cell lines, and in vitro antiviral activity against herpes simplex virus type 1. Because of the pharmacological potential and the highly diverse as well as complex architecture of these natural products, they have emerged as influential targets for total synthesis. Herein, we proposed to develop the short route entry to this class of alkaloids:
Objectives
  • A unified strategy will be investigated for the synthesis of various bioactive tetrahydroquinoline and indole-based diterpene alkaloids.
  • The main emphasis of the synthesis is the discovery of new reaction-based strategies to maintain step and atom economic pathways.
  • Some strategic organic transformations such as vinylogous ester synthesis, cross-coupling reaction, dearomatization reaction, and α-amination of ketone will be followed to complete the target molecules.
  • Detailed mechanistic investigation thorough structural analysis using NMR studies, IR spectroscopy, mass spectrometry, and density functional theory (DFT) computations will also be carried out to elaborate on the origin of reactivity and selectivity for the key reactions.
Nature of work
This project's initial part would be synthesizing the potential key intermediate. Later on, that intermediate will be applied for the synthesis of various tetrahydroquinoloine and indole-based diterpene alkaloids. A strategy-based multistep organic synthetic protocol will be followed to complete the target molecules.
Essential and Desirable Qualifications
  • The candidate should have a Master's degree (with a minimum of 55% marks or equivalent grade) in Chemistry from a UGC-recognized University or Institution.

Fellowship
As per DST norms, the fellowship is ?31,000/- p.m. plus HRA.
