Technical Assistant (m/w/d)
in Single Cell Omics & TCR Validation
Full Time - Mainz
We are looking for support in our Clinical Cancer Genomics unit to start as soon as possible. The successful
candidate will be working with patient biopsies, cell lines and murine tissue samples to support the
establishment and apply state-of-the-art single cell sequencing methods and immunoassays to contribute to
the development of novel immunotherapies against cancer and other diseases.
Your tasks and responsibilities: 
Processing including dissociation of fresh, thawed or fixed healthy or patient tissues 
A good performance of routine cell culture work including maintenance and expansion of mammalian
primary cells (e. g. 2D and 3D tumor cell lines and TIL expansion) 
Analysing of flow cytometry and preparation of cell suspensions for sorting 
Preparation of single cell or nuclei suspensions for NGS-based single cell sequencing (sc RNA-seq, sc TCR-
seq and multiomics) using the 10x Genomics platform 
Generation of c DNA and ATAC libraries and preparation for sequencing on the Illumina platforms (Mi Seq
or Nova Seq 6000) 
Performance of functional TCR validation assays (e.g. ELISpot and killing assays) and immunogenic antigen
screens 
Training of new employees, supervision of automated laboratory processes and writing standard operating
procedures (SOPs) 
Timely generation of high-quality data and communication of results to the team
What you bring: 
A completed professional training as BTA/MTA, biological laboratory assistant, or a Bachelor (B.Sc.)/Master
(M.Sc.) degree in natural sciences 
Experience in molecular biology techniques (RNA extraction, PCR, NGS library preparation) is required 
Experience in mammalian cell culture is required 
Previous professional experience would be desirable 
Experience in immunological techniques (e.g. ELISA, flow cytometry) would be of advantage 
Any practice in processing primary samples (patient material, mouse samples) would be of advantage 
Strong visual, verbal, and written communication skills in English and German as well as strong sense of
responsibility and high reliability are essential
